How to Read a Peptide Certificate of Analysis

A field-by-field framework for checking whether a peptide Certificate of Analysis is internally consistent, method-linked and traceable to the supplied batch.

Start With Document Identity

A Certificate of Analysis, or COA, should identify the material before it reports a result. Check the peptide name or sequence, batch number, document number, issue date and specification version against the quotation, label and packing records.

Revision status matters because a corrected COA can supersede an earlier copy. A controlled document normally shows who approved it and when, while unexplained edits, mismatched dates or cropped identifiers weaken traceability.

  • Match the batch number character for character across all supplied records.
  • Confirm that sequence, modification state and counterion description are not ambiguous.
  • Ask whether the COA is final, provisional or reissued.

Separate Specifications From Results

A specification states the acceptance criterion; a result states what the laboratory observed. Buyers should expect both, together with units and a clear pass or fail decision, rather than a result presented without its agreed threshold.

Identity, chromatographic purity, water, residual solvents and peptide content answer different questions. A single high purity percentage cannot substitute for the omitted tests or establish total composition.

Follow Each Number to Its Method

Analytical values are interpretable only in the context of a method. For HPLC, look for column, mobile phases, gradient, detection wavelength, injection details and integration rules; for mass spectrometry, look for ionization mode and expected mass basis.

The COA may summarize rather than reproduce a full method, but it should point to a method identifier or accompany raw-output extracts. Compare sample name and acquisition date on those outputs with the COA batch and approval timeline.

Perform an Internal Consistency Check

Read the COA as a connected data set. Molecular formula, theoretical mass, observed ions, sequence length, purity result and content basis should tell a compatible story; discrepancies deserve clarification before procurement decisions.

Record questions in a review log and request corrected controlled records rather than accepting informal explanations alone. A credible response distinguishes a clerical correction from retesting and preserves the history of both.

  • Flag impossible dates, reused chromatograms and inconsistent sample identifiers.
  • Recalculate simple mass-to-charge relationships where charge assignments are shown.
  • Check whether results are reported as-is, dry basis, anhydrous basis or salt-free basis.

Limitations / What this does not prove

  • A COA does not by itself prove that the supplied container matches the tested sample.
  • A passing result does not establish suitability for every downstream research method.
  • Document appearance or signatures alone do not prove laboratory competence.
  • A COA is not evidence of safety, efficacy or suitability for human or veterinary use.

FAQ

Is a COA without raw data automatically invalid?

Not automatically, but it provides less reviewable evidence. Buyers can request representative chromatograms, spectra or method references appropriate to the transaction.

Should HPLC purity and assay have the same value?

No. HPLC area purity describes relative detected peaks under one method, while assay or content aims to quantify target peptide on a stated basis.

What should happen when a COA is corrected?

The revised document should be identifiable, dated, approved and linked to the superseded version, with the reason for correction available.
